摘    要:[目的]诱导虎杖叶的愈伤组织并测定其中白藜芦醇的含量。[方法]以虎杖叶为组织培养材料,在28℃和黑暗培养条件下,在含有浓度0.2 mg/L 2,4-二氯苯氧乙酸(2,4-D)和浓度1 mg/L 6-苄氨基腺嘌呤(6-BA)的MS诱导培养基中进行诱导,15 d后把诱导出的愈伤组织转接到相同培养基中培养15 d,转接3次后,把愈伤组织切下放置在相同培养基上继续培养3次后,采用HPLC法测定虎杖叶愈伤组织中白藜芦醇的含量。[结果]诱导出的愈伤组织生长状况良好,并且测得虎杖叶愈伤组织中白藜芦醇的含量为0.24 mg/g。[结论]由叶诱导出的愈伤组织中含有白藜芦醇成分,用HPLC法测定其含量,方法简便、迅捷,结果可靠。

Abstract:[Objective] To induce the callus from P.cuspidatum leaves and to determine the content of resveratrol.[Method]P.cuspidatum leaves as tissue culture materials were induced on MS induction medium containing 0.2 mg/L 2,4-dichlorophenoxyacetic acid(2,4-D) and 1 mg/L 6-amino-benzyl adenine(6-BA) at 28 ℃ and dark.15 d later,the induced callus was transferred to the same culture medium for 15 d,which were transferred 3 times.Subsequently,the callus was cut and then cultured in the same medium for three times.The content of resveratrol in the callus induced fromP.cuspidatum leaves was determined by HPLC.[Result] The callus induced from P.cuspidatum leaves grew well,and its resveratrol content was determined at 0.24 mg/g.[Conclusion] The HPLC method was simple.Convenient,quick and accurate to determine the content of resveratrol in P.cuspidatum leaves.
