摘    要:[目的]研究测定茶枝柑皮中多糖的组成成分。[方法]以1-苯基-3-甲基-5-吡唑啉酮(PMP)进行柱前衍生化,采用反相高效液相色谱法,测定茶枝柑皮多糖中单糖衍生物。色谱条件为:色谱柱为Phenomenex Luna C18(4.6 mm×250 mm,5μm);流动相为浓度0.05mol/L混合磷酸盐溶液-乙腈(80∶20,V/V,pH=7.1);检测波长为245 nm。[结果]茶枝柑皮多糖由甘露糖、核糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、木糖、阿拉伯糖组成,其摩尔比为0.08∶0.002∶0.05∶4.80∶1∶1.52∶0.11∶3.82。[结论]该方法具有操作简单、快速、重复性好、回收率高等特点,可用于柑橘果胶多糖中单糖组成及含量的测定。

Analysis of Polysaccharides Composition of Citrus reticulata ' Chachi' Peel

Abstract:[Objective] Polysaccharides composition of Citrus reticulata 'Chachi' peel was studied.[Method] Taking PMP for pre-column derivatization,monosaccharide derivatives in polysaccharides of Citrus reticulata 'Chachi' peel were detected by Reversed-Phase High Performance Liqnid Chromatography(RP-HPLC).Chromatographic conditions were Phenomenex Luna C18(4.6 mm×250 mm,5 um),0.05 mol/L phosphate solution – acetonitrile(80∶20,V/V,pH=7.1) as mobile phase,and detection wavelength at 245 nm.[Result] Polysaccharides of Citrus reticulata 'Chachi' peel were composed of mannose,ribose,rhamnose,galacturonic acid,glucose,galactose,xylose,and arabinose.Their molar ratio was 0.08:0.002:0.05:4.80:1:1.52:0.11:3.82.[Conclusion] This method was simple,rapid,repeatable and has high recovery rate.Thus,it could be used for the determination of monosaccharide composition and content in citrus pectin polysaccharides.
