大豆DNA导入引起稻米蛋白质含量变异的遗传稳定性及赖氨酸含量分析

摘    要:利用浸种及苗期浇灌法和花粉管通道法将高蛋白含量的大豆总DNA导入水稻,提高了水稻后代种子的蛋白质含量和总氨基酸及赖氨酸含量。三代水稻种子蛋白质含量的数据分析结果表明:利用大豆总NDA的导入不仅可以迅速有效地提高稻米蛋白质和赖氨酸含量,而且稻米高蛋白、高忱酸变异性状还能够稳定表达至第三代;本试验还分析比较了两种DNA直接导入法,虽然从总体而言,在提高种子蛋白质含量以及保持后代种子高蛋白性状的遗传稳定性方法,利用花粉管通道法导入外源大豆DNA,相对浸种及苗期浇灌法效果更好,但从不同株系蛋白质含量的追踪分析结果显示,连续两年采用大豆DNA溶液浸种及苗期浇灌处理获得的株系,在维持后代种子高蛋白性状的表现方面效果更明显:三代经大豆DNA处理的水稻种子,赖氨酸含量都伴随着氨基酸总含量的提高而提高,而且第二代和第三代种子的赖氨酸含量都超过0.5%。

Abstract:The exogenous DNA from soybean was introduced into rice seeds by soaking seeds, moreover irrigating seedlings, and through the pollen tube pathway after self pollination. The protein contents and lysine contents in three generation seeds were analyzed. The results indicated: The average contents of protein in three generation contrasts' seeds were 12.91%?13.05% and 13.11%.The average contents of protein in three generation seeds treated with soybean DNA by soaking seeds, moreover irrigating seedlings were 16.64%?17.30% and 16.96%.The average contents of protein in three generation seeds induced into soybean DNA through the pollen tube pathway were 1987%?17.03% and 17.05%. The average contens of protein in three generation seeds introduced into soybean DNA were increased distinctly over their contrasts;The effectiveness of two kinds of ways to introduce exogenous DNA was compared. The way to introduce exogenous DNA through the pollen tube pathway was better than the way to introduce exogenous DNA once by soaking seeds, moreover irrigating seedlings in increasing seed protein contents. But it was not as good as treating with soybean DNA two times by soaking seeds, moreover irrigating seedlings; Lysine contents followed all of amino acid contents in three generation seeds. Lysine contents in the second and third generation seeds treated were also over 0.5%.
