抗苯并咪唑类杀菌剂的香蕉潜伏炭疽菌β—微管蛋白DNA测序

摘    要:将经特克多诱导后获得抗性的2个Colletotrichum musae菌系,及其相应的示经任何药剂处理的敏感菌系的DNA作为模板,用真菌β-微管蛋白基因的2个引物进行PCR扩增,均得到一段长874bp的β-微管蛋白的编码序列,并含有1个53bp的内含子。通过PC/Gene软件分析,与已报道的其它5种真菌β-微管蛋白基因相比,核苷酸同源性最高达97.8%,成氨基酸同源性最高达99.3%;抗性菌系与敏感菌系相比,在198和200位位置的氨基酸并没有发生突变,但在186、232、313和329位置氨基酸发生了变异。

Abstract:The total DNA isolated from TBZ-resistant and sensitive isolates of C.musae was used as templates in PCR amplification. The PCR product was cloned into T-plasmid and then sequenced. Homology comparison using PC/Gene software showed that the sequence of this fragment had a high homology to the sequences of β-tubulin genes of other 5 fungi reported earlier. TBZ-resistant isolates did not carry the allelic mutation at amino acid codes 198 and 200 of β-tubulin gene when compared to the sensitive isolates. But the amino acid changed in codens 186 232 313 and 328 of this fragment.
