铜藻膳食纤维提取条件的研究

摘    要:以铜藻为原料,采用化学与酶解结合的方法,经酶解、碱提取、沉淀、漂白、活化、烘干等工艺处理提取膳食纤维,研究了酶解、碱提取、漂白工艺条件对铜藻膳食纤维提取的影响,用正交设计法筛选出铜藻膳食纤维提取的最优工艺条件。由正交试验结果分析可知,酶解工艺的最优条件为:纤维素酶用量90U/g、木瓜蛋白酶用量5000U/g、酶解时间1.5h;碱提取工艺的最优条件为:20倍的200g/LNa2CO3溶液、处理时间2h、处理温度85℃;漂白工艺的最优条件为:3倍的0.3%NaClO溶液、DH7、漂白时间40min。结果表明:在该工艺条件下提取的铜藻膳食纤维的产率为35.4%,颜色较白,总膳食纤维干基含量为78.6%,膨胀力为85.8mLk,持水力为4220.0%,蛋白质含量0.45%,总灰分含量为18.3%。该方法所提取的铜藻膳食纤维的产率较高。

Study on Optimization of Dietary Fiber Extraction from Sargassu horned

Abstract:The optimal process conditions of dietary fiber extraction from Sargassu horneri were studied by means of enzymatic and chemical method. The optimum technological condition of extraction of dietary fiber have been chosen from Sargassu horrteri by orthogonal design method after studying the influence of craft con- ditions such as enzymolysis, the alkali withdraws, bleaching on dietary fiber extraction from Sargassu horrteri. From the results of the orthogonal design,we knew that the optimum technological condition of enzymolysis was 90 U/g of the cellulose use level, 5 000 U/g of the papain use level, 1.5 h of the enzymolysis time, the optimum technological condition of the alkali withdraws was 20 times of the Na2CO3 use level, 2 h of the processing time, 85 ℃ of the processing temperature. The optimum technological condition of bleaching was 0.3% of NaClO concentration, 3 times of the NaCIO use level, pH 7, 40 min of the bleaching time. The results at opti-real conditions for extracting Sargassum DF were 35.4 % of yield, nearly white in color, 78.6% of DF contents, 85.8 mL/g of expansion capacity and 4 220.0% of water holding capacity, 0.45% of protein content, and 18.3 of total ash content.
