潮间带菌株CZⅣ-1068的鉴定及抑菌活性研究

摘    要:CZⅣ-1068是从舟山长峙岛潮间带海泥中分离到的一株细菌,最适NaCl浓度范围为1%~3.5%,经琼脂扩散法抗菌模型筛选表明对大肠杆菌、副溶血性弧菌、枯草芽孢杆菌、金黄色葡萄球菌、藤黄微球菌和白假丝酵母有拮抗作用。利用形态学观察、生理生化反应对菌株进行了初步研究,并利用PCR的方法扩增了CZⅣ-1068的16S rDNA,然后进行核苷酸序列测定分析,结果表明CZⅣ-1068与Bacillus sp.82344(AF227848)相似性高达100%,初步确定为腊状芽孢杆菌的一种。

Identification and Antimicrobial Activity of Strain CZIV-1068 from Sediment of Intertidal Zone

Abstract:CZⅣ-1068, which was isolated from sediments of intertidal zone in Changzhi island, is weekly halophilic bacterium and its optimum range of salt concentration is 1%-3.5% of NaCl. It was found that the metabolities of the strain had antagonism on Escherichia coli, Vibrio parahaemolyticus, Bacillus subtilis, Staphylococcus aureus, Micrococcus luteus and Candida albicans by using agar diffusion method. CZⅣ-1068 was classified into Bacillus cereus subgroup based on morphology, analysis of physiology and biochemistry and molecular biology methods. The sequence of 16S rDNA from CZⅣ-1068 strain was similar to Bacillus sp. 82344 (AF227848) compared with those in database. The evolutional distance was calculated and phylogenetic tree was drawn based on the sequence data.
