摘 要: | 分析了不同外值体、培养基类型和pH值对雷公藤愈伤组织诱导的影响,并进一步研究了雷公藤愈伤组织的生理生化特征以及内酯醇在愈伤组织中的积累。研究结果表明,雷公藤愈伤组织诱导的最佳外植体为组培苗的嫩叶,最佳培养基为MS,最佳pH值范围为5.8-6.1。愈伤组织在生长周期的中期(第4周)和末期(第8周), SOD和POD活性最高, MDA含量和PAL活性则较低;在生长稳定期, SOD和POD活性较低,而MDA含量和PAL活性则较高。对于不同外植体愈伤组织中的雷公藤内酯醇含量,根愈伤与叶愈伤相近,均明显高于茎愈伤;愈伤组织在生长周期的后期(第6-8周)内酯醇的积累更高。

Growth traits and triptolide accumulation of Tripterygium wilfordii callus |

Abstract: | This study investigated the effects of different explants, culture media and pH on induction of Tripterygium wilfordii callus. And then physiological and biochemical properties and triptolide accumulation in the callus were further analyzed. The results indicated that, to induce T. wilfordii callus, the best explants were tender leaves of tissue culture seedlings, the most suitable medium was MS, and the optimal pH values ranged from 5.8 to 6.1. The activities of SOD and POD both reached the maximum at the middle stage (4th week) and the end (8th week) of the callus growth period, when MDA content and PAL activity showed relatively low values. At the stationary growth phase, SOD and POD had low activities, while MDA content and PAL activity were high. As to triptolide content in the callus of different explants, the root callus and the leaf callus shared similar levels, significantly higher than the stem callus. The callus produced higher triptolide amount at the latter stage ( 6th to 8th week) of the growth period. |
