摘    要:本研究从金钱树(Zamioculcas zamiifolia)白绢病(Southern blight)病株上分离到一株产纤维素酶菌株SM,经形态观察和rDNA—ITS序列分析鉴定为齐整小核菌(Sclerotium rolfsii Sacc),经以羧甲基纤维素钠为唯一碳源的培养基培养和刚果红染色测定,证明SM菌株可产生高活性纤维素降解酶。本文对SM菌株产纤维素降解酶的液态发酵条件进行了研究,结果表明:在起始pH6.0,无机氮:有机氮:纤维素碳源之比为0.07g:1.4g:1.6g,温度为30℃,摇床转速为160r/min,发酵培养时间为5d的条件下,该菌株发酵液的CMC酶活性达到11.7U/mL,滤纸酶活性达到2.156U/mL,β-葡萄糖苷酶活性达到3.911U/mL。

Isolation and Characteristic Identification of a Strain Which was Isolated from the Southern Blight Plants of Zamioculcas zamiifolia to Yield Cellulase Enzyme

Abstract:An strain SM which can produce cellulose was isolated from the southern blight diseased plants of Zamioculcas zamiifolia and then was identified as Sclerotium rolfsii Sacc using morphologic observation method and rDNA-ITS sequence analysis method. Through cultivating on substrate with exclusive carbon source-CMC-Na, the strain SM was proved to be able to produce high activity cellulose using congo red staining test. The liquid fermentation conditions of the strain SM producing cellulolytic enzymes were optimized, and the results showed that under the condition that initial pH 6.0, inorganic nitrogen: organic nitrogen: cellulose carbon source of it is 0.07 g: 1.4 g: 1.6 g, culture temperature is 30℃, rotation speed is 160 r/min, and the cultivated days is 5 d, the fermentation liquid of strain SM shows the optimal levels of CMC enzyme, filter paper enzyme and β-glycosidase activity which is 11.7 U/mL, 2.156 U/mL and 3.911 U/mL, respectively.
