微环境调控对半夏无糖组培苗根、叶显微结构的影响

摘    要: 以半夏组培苗为研究材料,应用植物无糖组培快繁技术,研究探讨了微环境控制对半夏无糖组培苗根、叶显微结构的影响。研究结果表明,半夏无糖组培苗根、叶显微结构与有糖组培苗(CK)相比其保水能力具明显优势。半夏无糖组培苗叶片具有发育较好、较厚的栅栏组织和海棉组织,细胞排列紧密有序,中脉发达,且叶表皮具有蜡质层;而半夏同龄有糖组培苗叶片的栅栏组织和海棉组织细胞发育较差,中脉及其维管束发育不完全。半夏无糖组培苗根部细胞排列紧密组织分区明显,初生组织和次生组织已分化;能明显观察到疏导组织、髓射线和凯氏带;半夏有糖组培苗根生长发育不良,初生组织和次生组织分化不明显,疏导组织不发达。

Abstract:In this research, plantlet culture of Pinellia ternate was used as material to study the effect of different light intensities, photoperiods, hormone concentrations on the photophysiology and microstructure of leaf and root of plantlet in sugar-free tissue culture of Pinellia ternate(Thunb.)Breit..The result showed the microstructure of P.ternate of leaf and root had obvious difference between the plantlet cultured in the sugar-free condition and in the sugar (CK) condition.The plantlet developed better under the sugar-free condition than CK.It had thick palisade tissue, thick sponge tissue, thick epicutile wax layer, stronger midrib and more stomata in lower epidermis.But the plantlet culture under the sugar condition had thickless palisade tissue, thickless sponge tissue, weaker midrib and without epicutile wax layer on epidermis.So the plantlet culture under the sugar-free condition had stronger protective tissue to hold water.The cells of root of plantlet of P.ternate under the condition of sugar-free culture were tidy and tightly secondary tissue.And the tissue system of the root was obvious.Its primary tissue and secondary tissue were differentiation.Conducting tissue, pithray was significantly obvious.But the root of plantlet of P.ternate under the condition of sugar culture developed badly.Its tissue system was not differentiation and its conducting tissue developed weaker.〖WTHZ〗Key words 〖WT5BZ〗 ZK(]Pinellia ternate(Thunb.)Breit.; sugar-free tissue culture; paraffin section; scanning electron microscope; microstructure
