江苏8个克氏原螯虾群体遗传多样性微卫星分析

摘    要:利用8个微卫星标记分析了江苏省8个地区(大浦、平望、漆塘、官滩、西顺河、太平、固城、洋河滩)克氏原螯虾的遗传多样性。研究表明:8个群体全部64个位点经Bonferroni法校正后均显著偏离Hardy-Weinberg平衡且绝大多数位点显示杂合不足。8个群体中大浦群体遗传多样性水平最高,平望群体遗传多样性水平最低。群体间遗传分化指数FST及AMOVA分析表明,8个群体遗传分化指数为0.126,存在中等程度的分化(0.05FST0.15)。基于遗传距离构建的UPGMA聚类树显示,8个群体可分为4组:平望群体、漆塘群体和大浦群体聚为一组,太平群体、西顺河群体与官滩群体聚为一组,而固城群体、洋河滩群体分别自成一组。

Abstract:Genetic diversity of eight Procambarus clarkii stocks in Jiangsu Province (Dapu, Pingwang, Qitang, Guantan, Xishunhe, Taiping, Guchen, Yanghetan) were investigated using 8 microsatellite DNA loci. The result showed that all the eight loci were highly or medium polymorphic. Tests of departures from Hardy-Weinberg equilibrium indicated all 64 loci in 8 stocks was significant deviation from Hardy-Weinberg equilibrium after Bonferroni correction and most of them has significant heterozygosity deficiency. All of the 8 stocks showed higher genetic diversity, and genetic diversity of the stocks in DP was the higher, that of PW were lower. FST and AMOVA analysis across all stocks and loci indicated that there was medium level of divergence among 8 stocks, FST= 0.1268 (0.05
