紫山药花青素提取工艺及抗肿瘤活性筛选

摘    要:目的研究玫瑰花青素抗疲劳和抗氧化功效。方法通过灌胃给予健康雄性昆明小鼠低、中、高剂量(依次为100、150和200 mg/kg)玫瑰花青素,30 d后负重游泳;测定小鼠负重游泳时间、血清和肝脏中尿素氮(BUN)、血乳酸(BLA)和丙二醛(MDA)含量、乳酸脱氢酶(LDH)、谷胱甘肽过氧化物酶(GSH-Px)和总超氧化物歧化酶(T-SOD)活性以及总抗氧化能力(T-AOC)等生化指标;根据线性方程分析玫瑰花青素总抗氧化能力与其抗疲劳功效的关系。结果与对照组相比,低、中、高剂量玫瑰花青素使小鼠负重游泳时间极显著增加(P<0.01);低、中、高剂量组小鼠血液中BLA和BUN含量显著降低(P<0.05或P<0.01),LDH活性极显著增强(P<0.01);低、中、高剂量组小鼠血清和肝脏中的MDA含量均降低,GSH-Px和T-SOD活性均显著增加(P<0.05或P<0.01),T-AOC显著提高(P<0.05或P<0.01)。相关性分析表明:随着小鼠血液中T-AOC增加,BLA和BUN含量降低,LDH活性升高。结论玫瑰花青素能有效增加小鼠体内抗氧化能力,缓解疲劳;抗疲劳和抗氧化功效与玫瑰花青素提取物灌胃剂量存在依赖关系。

Abstract:PurposeTo study the anti-oxidation and anti-fatigue effects of rose anthocyanins. MethodsMale healthy Kunming mice were given low (100 mg/kg), medium (150 mg/kg) and high (200 mg/kg) dose rose anthocyanins for 30 days. After weight-bearing swimming, the time of weight-bearing swimming was measured, and the biochemical indexes in serum and liver such as the contents of blood urea nitrogen (BUN), blood lactic acid (BLA) and malondialdehyde (MDA), the activities of lactic dehydrogenase (LDH), glutathione peroxide dismutase (GSH-Px) and total superoxide dismutase (T-SOD), and total anti-oxidant capacity (T-AOC) were measured. According to the linear equation, the relationship between anti-oxidant capacity and anti-fatigue effect of rose anthocyanins was analyzed. ResultsCompared with the control group, low, medium and high dose of rose anthocyanins extremely significantly prolonged the swimming time of mice with load (P<0.01); the contents of BLA and BUN in the blood of mice with low, medium and high dose of rose anthocyanins significantly decreased (P<0.05 or P<0.01), but the activity of LDH extremely significantly increased (P<0.01); the contents of MDA in the serum and liver of mice with low, medium and high dose of rose anthocyanins decreased, the activities of GSH-Px and T-SOD increased significantly (P<0.05, P<0.01), the T-AOC significantly increased (P<0.05, P<0.01). The correlation analysis showed that with the increase of T-AOC in mice, the contents of BLA and BUN decreased, and the activity of LDH increased. ConclusionRose anthocyanins can effectively increase the anti-oxidation ability and relieve fatigue, and the anti-oxidation and anti-fatigue effects are dependent on the dose of gavage.
