测定生姜蛋白酶活性的新方法

摘    要:通过对生姜蛋白酶进行分离纯化,并将其加热使酶失活作为参照,用考马斯亮蓝染色法测定蛋白酶活力。研究了pH值、温度、EDTA和金属离子(Na+,Mg2+,Cu2+,Ca2+)等因素对生姜蛋白酶活性的影响。结果表明,以失活的生姜蛋白酶为参照,可以简洁的测定出生姜蛋白酶的活性,与已往测定生姜蛋白酶活性相比较,具有方法简单,消除腐蚀危害,可望简洁而环保的测定出生姜蛋白酶的活性。

Abstract:In this paper,a new method for detecting ginger protease activity was proposed by using inactive ginger protease as a reference.Coomassie brilliant blue staining method was employed to detect the activity of ginger protease.The influence of some factors,such as the pH,temperature,metallic ions(Na+,Mg2+,Cu2+,Ca2+)and EDTA on the activity of the protease was discussed.The results indicate that taking the inactive ginger protease as a reference could detect the activity of ginger protease laconically,which exh...
