Abstract:To explore the effects of different selenium sources and concentrations on seed germination and material transformation of Medicago sativa‘Pianguan’.This study used Medicago sativa‘Pianguan’seeds as materials,using different concentrations of sodium selenite(0,0.3,0.6,1.2,2.4,4.8,9.6 mg·L-1)and selenium methionine(0,1,5,10,20,40 mg·L-1)germination test in an artificial climate incubator.The result showed:0.3 mg·L-1 sodium selenite and 1 mg·L-1 methionine selenium significantly promoted the hypocotyl elongation of alfalfa seedlings.The hypocotyl diameter of alfalfa seedlings increased significantly under 1 and 5 mg·L-1 methionine selenium treatment.The content of reducing sugars increased significantly when the concentrations of sodium selenite and selenium methionine were 0.3,0.6 mg·L-1 and 5,10 mg·L-1 respectively.Except for 4.8 mg·L-1 sodium selenite and 20,40 mg·L-1 selenium methionine,the activity of nitrate reductase can be significantly improved at each selenium concentration,0.3 mg·L-1 sodium selenite and 40 mg·L-1 methionine selenium can significantly increase the total amino acid content of alfalfa seedlings.Alfalfa seed was more sensitive to sodium selenite during the germination process,while their sensitivity to selenium methionine was relatively weak.This is more conducive to the later morphogenesis.Comprehensive analysis shows that 5 mg·L-1 methionine selenium is an ideal growth promoter and selenium biological fortifier at the germination stage of alfalfa seeds.
