| 摘 要： | 在CTAB法及SDS法的基础上设计了6种提取方法,比较了不同方法对神秘果幼嫩叶片DNA的提取效果。结果表明:改进的SDS法,通过提高提取缓冲液中重要成分的含量,与二次纯化的方法相结合,提取的DNA无明显降解、杂质少,OD260/OD280值为1.62,接近标准值,是有效提取神秘果基因组DNA的方法。

| Abstract: | Genomic DNA of leaves of mysterious fruit was extracted by means of six different methods of CTAB,SDS and all kinds of improved methods,The extraction DNA of these methods has been compared.The result showed that:the SDS improve method,increased the important content of extraction buffer and used method of two purification it was an effective way to solve all kinds of problem,degradation was little.impurity was little.That was the best method since the DNA extracted by it had the best OD260/OD280 ratio,it was 1.62 or so. |
