双孢蘑菇胞外分泌蛋白质组的功能预测及分析

摘    要:以双孢蘑菇(Agaricus bisporus)10448个蛋白为研究对象,综合利用信号肽分析软件SignalP 4.1、跨膜结构预测软件TMHMM v2.0、GPI锚定位点预测软件big-Pi Predictor以及亚细胞蛋白定位软件TargetP1.1等生物信息学软件,筛选双孢蘑菇胞外分泌蛋白质序列,使用分析软件BLAST2GO进行功能注释。筛选得到469个胞外分泌蛋白质序列,统计发现编码这些蛋白质的开放阅读框(open reading frame,ORF)最长为4800bp,平均为1233bp。除去假定蛋白质,双孢蘑菇胞外分泌蛋白质的生物代谢途径主要有生物大分子代谢(22.2%)、碳水化合物代谢(17.4%)、脂质代谢(3.2%)、芳香化合物代谢(2.2%)等。同时,共发现133种酶类,其中包括糖苷水解酶(38.5%)、多铜氧化酶(7.4%)、肽酶(7.0%)、果胶裂解酶(4.0%)和羧酸酯酶(2.9%)等。

Abstract:Using informatics softwares including signal peptide analysis software SignalP 4.1,transmembrane domain prediction software TMHMM v2.0,GPI anchorage predition software big-pi Predictor and subcellular protein location software TargetP 1.1,469 secretory proteins were identified out of from totally 10448 proteins of Agaricus bisporus from NCBI (National Center for Biotechnology Information).The ORFs (open reading frame) of the 469 proteins were found to be 4800 bp in maximum,and 1233 bp in average.Their functions were further annotated by protein analysis software BLAST2GO.Excluding hypothetical proteins,major biological pathways of these identified proteins werc mainly associated with metabolic pathways of biological macromolecules (22.2%),carbohydrates (17.4%),lipids (3.2%) and aromatic compounds (2.2%).In addition,133 enzymes were identified,of which 38.5% were glycoside hydrolases,7.4% were multicopper oxidases,7.0% were peptidases,4.0% were pectatelyases and 2.9% were carboxylesterases.
