摘    要:为研究不同乳脂率德宏奶水牛乳腺组织中FATP1基因的表达情况,试验选取216头处于泌乳盛期的健康德宏奶水牛,分为高乳脂率组(H组)、中乳脂率组(M组)和低乳脂率组(L组),采集乳样,用乳品分析仪测定乳脂率,利用气相色谱法测定脂肪酸含量。采集乳腺组织,利用荧光定量PCR法检测FATP1基因的mRNA表达水平,并与乳脂率和脂肪酸含量进行相关性分析。结果表明:H组乳脂率、总脂肪酸含量和长链脂肪酸含量均极显著高于M组和L组(P<0.01),M组显著高于L组(P<0.05)。不饱和脂肪酸含量H组极显著高于L组(P<0.01),显著高于M组(P<0.05),而M组与L组无显著差异(P>0.05)。FATP1基因mRNA表达水平为H组极显著高于L组(P<0.01),而H组与M组、M组与L组均无显著差异(P>0.05)。相关性分析表明,乳脂率与乳中总脂肪酸、长链脂肪酸和不饱和脂肪酸含量间呈极显著正相关(P<0.01),FATP1基因mRNA表达量与乳脂率、脂肪酸总量、长链脂肪酸含量、不饱和脂肪酸含量均呈极显著正相关(P<0.01)。研究表明在乳脂合成的过程当中FATP1基因对脂肪酸的摄取和转运起到重要的调控作用。

Abstract:In order to study the expression of FATP1 gene in mammary tissues of Dehong dairy buffalo with different milk fat contents, 216 healthy Dehong milk buffaloes in the high milk fat contents group(H group), medium milk fat contents group(M group) and low milk fat contents group(L group) were selected in the experiment. There were three buffaloes in each group. All animals were sampled at peak of lactation. Milk was collected and milk fat content were tested by milk composition analyzer. Fatty acid contents were measured by gas chromatographic. The expression level of FATP1 in mammary tissue were measured by real-time PCR. The correlation between the FATP1 gene expression and milk fat,fatty acid content in buffalo milk were analyzed. The results showed that the milk fat contents, total fatty acid and long chain fatty acid contents(LCFA) were extremely significantly higher in group H compare to group M and group L(P<0.01), group M was significantly higher than group L(P<0.05). The content of unsaturated fatty acid(UFA) in group H was extremely significantly higher than group L(P<0.01), and group H was significantly higher than that in group M(P<0.05), but there was no significant difference between group M and group L(P>0.05). The expression level of FATP1 gene in group H were extremely significantly higher than group L(P<0.01), while there was no significant difference between group H and group M or group M and group L(P>0.05). Correlation analysis showed that the milk fat content and the contents of total fatty acids, LCFA and UFA in milk were extremely significantly positive correlation(P<0.01). The expression level of FATP1 m RNA was significantly positively correlated with milk fat content, total fatty acid, LCFA and UFA content(P<0.01). The results of this study indicate that the FATP1 gene plays an important role in regulating fatty acid uptake and transfer during synthesis of milk fat.
