27份核桃种质资源亲缘关系的ISSR分析

摘    要:采用ISSR分子标记技术对4个种的27份核桃种质进行亲缘关系分析。从120条ISSR引物中筛选出扩增条带清晰稳定、重复性好的10条引物对27份核桃种质基因组DNA进行扩增,共扩增出129条带,其中91条呈现多态性,多态性条带的比例平均为70.54%。不同核桃种质间遗传相似性系数在0.496 1~1.000 0之间。研究结果表明:27份核桃种质具有较为丰富的遗传多样性,铁核桃与核桃的亲缘关系较近(最近),与野核桃的亲缘关系最远。基于遗传相似性系数的UPGMA聚类结果表明,27份核桃种质按照种的划分被明显地分成4大组,与传统形态分类一致。该结果为核桃种质资源的进一步研究和利用提供参考。

Abstract:ISSR technique was applied to analyze the genetic relationships of 27 walnut germplasm resources from 4 species.Ten primers with excellent reproducibility and limpid bands were screened out from 120 ISSR primers to amplify the genome DNA of 27 germplasms.129 bands were amplified out totally,in which 91 were polymorphism bands,accounting for 70.54% of the total.The value of Nei’s genetic similarity index of 27 germplasm varied from 0.496 1 to 1.000 0.The results demonstrated a relatively rich genetic diversity of the germplasm and genetic relationship of the samples.Cluster analysis with UPGMA method showed that the 27 germplasm could be divided into 4 groups,which was consistent to the morphological classification.The study will provide references for further development and utilization of walnut germplasm.
