Abstract:The Notch signaling and TLR signaling can interact cooperatively with each other to regulate the production of inflammatory factors and activate the macrophages.However,it is little known about how the Notch signaling regulates the inflammatory responses in porcine alveolar macrophages (PAMs).In this study,a model study was established in PAMs for inflammatory responses induced by LPS/TLR4 in order to investigate effect of LPS treatment on Notch signaling and regulation of LPS-induced inflammatory responses by Notch signaling.The results showed that LPS treatment increased the expression of ligands Jagged1,Jagged2 and Dll4,and up-regulated the expression of downstream target gene Hey1.Furthermore,the Notch signaling regulated LPS-induced inflammatory responses as demonstrated using DAPT treatment and silence RBP-J expression.These findings revealed that the Notch signaling in PAM regulated the LPS-induced inflammatory responses and paved the way for investigation of the Notch signaling in pig-associated bacterial infections.
