犬冠状病毒胶体金检测卡制备与评价

为快速检测犬冠状病毒,研究利用胶体金免疫层析技术制备和评价犬冠状病毒抗原检测卡。试验通过将胶体金标记的CCV Ab1和鼠IgG涂布于聚酯纤维膜上作为标记垫,生物素耦联的CCV Ab2处理在样品垫上,链霉亲和素和羊抗鼠IgG包被于NC膜上分别为检测线和质控线,与吸水纸和PVC底板制备犬冠状病毒抗原胶体金检测卡,并对其评价。结果:该检测卡不与其他病毒发生交叉反应,可检测浓度低至2 ng/mL的样本,24月是稳定的。结论:该测试板具有良好的特异性、灵敏度、重复性和稳定性,并且与RT-qPCR的结果较符合。因此,该检测卡能快速、准确地检测犬冠状病毒,为动物临床诊断和治疗提供参考。

Preparation and Evaluation of Canine Coronavirus Colloidal Gold Detection Card

For rapid detection of canine coronavirus,in this study,gold immunochromatographic assay(GICA)was used to prepare and evaluate canine coronavirus antigen detection card.The colloidal gold-labeled CCV Ab1 and mouse IgG were coated on Polyester fiber membrane as labeling pad,and the biotin-coupled CCV Ab2 was treated on the sample pad,the detection line and quality control line were coated on NC membrane by Streptavidin and Ovine anti-rat IgG respectively,and the colloidal gold detection card of Canine coronavirus antigen was prepared with absorbent paper and PVC substrate.Results:The test card did not cross-react with other viruses.It was stable in 24 months for samples with concentration as low as 2 ng/mL.Conclusion:The test plate has good specificity,sensitivity,repeatability and stability,and is in good agreement with RT-qPCR.Therefore,the card can detect canine coronavirus rapidly and accurately,and provide reference for animal clinical diagnosis and treatment.
