摘    要:通过研究摸索出适宜提取柑桔叶片总DNA的方法.利用异源DNA探针——小麦rDNA克隆pTA71,与用四种核酸内切酶EcoR 1、Pst 1、BamH 1及Bgl Ⅱ酶切消化的几种柑桔叶片总DNA杂交,结果表明,pTA71—EcoR 1、pTA71—Pst 1、pTA71—BamH 1、pTAT1—Bgl Ⅱ四种探针一酶组合均可在本地早、宜昌橙、枸橼和金柑等柑桔类型中检测到RFLP差异。利用rDNA的RFLP变异可以从分子水平上研究柑桔遗传多样性,探讨柑桔及其近缘属植物的起源、进化。

Abstract:A suitable method for total DNA extraction from citrus leaves was developed in this study. The total ge-nomic DNA digested with four restriction enzymes (EcoR I, Pst I , BamH I ,Bgl I ) was hybridized with a heterologous probe-pTA71 clone of wheat rDNA. Results showed that pTA71-EcoR I , pTA71-Pst I, pTA71-BamH I and pTA71 -Bgl I revealed some RFLP difference among Bendizao Tangerine (Citrus reticulata Blanco) .Ichang Papeda(C. ichangensis Swingle) and their F1 hybrid H10, Etrog Citron (C. medica L. ) and Kumquat (Fortunella crasstfolia Swingle). The RFLPs of rDNA in citrus could be used as molecular markers to assess the genetic diversity of citrus and its related genera and clarify their evolutionary relationships.
