摘    要:以来自中国热带地区的2个疫霉种(柑桔褐腐疫霉P.citrophthora和芋疫霉P.colocasiae)16个菌株的菌丝体的总DNA为模板,使用5种随机引物进行PCR扩增,获得随机扩增多态性DNA(RAPD),分析与各菌株相对应的RAPD的相似性。结果表明,RAPD不但能辨别这2个疫霉种间的差异,而且可以辨别种内的分类单元。初步肯定了RAPD可以用于中国热带地区疫霉菌的分类鉴定。

Abstract:Total DNAs were extracted from mycelia of 16 isolates from 2 species of Phytophthora (P. citrophthora and P. colocasiae) in the tropical area of China and used as templates for PCR amplification with 5 arbitrarily-chosen 10 -mer oligonucleoides as primers. The PCR products were separated in agarose gels, and the similarity of RAPD band patterns of the isolates were analyzed by using cluster analysis. Results showed that RAPD was able to identify not only the two Phytophthora species but the intraspecific taxons. It is preliminarily believed that RAPD can be used for taxonomic identification of Phytophthora in tropical China.
