川青牛胆离体繁殖技术优化

摘    要:为优化青牛胆离体繁殖技术,建立四川青牛胆快速繁殖技术体系的方法,本实验以川青牛胆幼嫩茎段为外植体,通过组织培养技术,探究适用于幼嫩茎段快速繁殖的最佳培养基。实验结果表明:最佳快速繁殖培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA,诱导率达90.0%;最佳增殖培养基为:MS+0.5 mg/L 6-BA+0.05 mg/L NAA;最佳生根培养基为:MS+1.0 mg/L 6-BA,生根率为97.5%;驯化移栽成活率为100.00%;本研究为川青牛胆优质种苗规范化生产和新品系选育提供了技术参考。

Abstract:In order to optimize the in vitro propagation technology and establish the method of rapid propagation technology system of Sichuan Tinospora sagittata, in this experiment, the young stem segment of Sichuan Tinospora sagittata was used as explants, and the optimal medium for rapid propagation of young stem segment was explored through tissue culture technology.The results showed that the optimal medium for rapid propagation was MS+1.0 mg/L 6-ba +0.1 mg/L NAA, and the induction rate reached 90.0%.The optimal culture medium was MS+0.5 mg/L 6-ba +0.05 mg/LNAA;The optimal rooting medium was MS+1.0 mg/L6-BA, and the rooting rate was 97.5%.The survival rate of acclimation and transplantation was 100.00%.This study provided technical reference for standardized production and new strain breeding of high quality Sichuan Tinospora sagittata.
