摘    要:目的]探索一株新的空多角体突变的HaSNPV的生物学性质。方法]利用同源重组的方法构建HaSNPV细菌人工染色体的过程中,发现一株多角体病毒对棉铃虫幼虫无毒性。通过电镜观察和Western-blot检测对多角体的生物学性质进行了初步探索。结果]细胞和多角体的电镜结果表明,该病毒能形成正常的多角体外壳,但是不能包装病毒粒子。通过PCR检测,发现其ORF51-54片段缺失,但这一多角体的突变并不与多角体基因和p10基因有关,ORF53(fp25)的回复也证明fp25的缺失不是造成此突变的原因。结论]该突变株将为HaSNPV基因功能的研究提供良好的技术平台与研究素材。

Abstract:Objective] The aim of the research was to discuss the biological characteristics of a newly-discovered strain of HaSNPV with polyhedrosis mutation.Method] A strain of polyhedrosis virus without toxicity to Helicoverpa armigera larvae was found during the construction process of bacterial artificial chromosome of HaSNPV by the method of homologous recombination.The biological characteristics of the polyhedrosis were preliminarily discussed by electron microscope observation and Western-blot detection.Result] The observation on polyhedrosis and the infected cells through electron microscope showed that the virus could form normal polyhedron coat,but it could not envelope any virion.PCR detection found that the virus was lack of ORF51-54 fragment,but this mutation wasn't related with polyhedrosis gene and p10 gene.The recovery of ORF53(fp25) also proved that the lack of fp25 was not the reason that caused this mutation.Conclusion] This mutant would provide a good technology platform and research materials for studying the functions of HaSNPV gene.
