刺龙牙组织培养与快繁技术的研究

摘    要:为加快刺龙牙的繁殖选优速度,进行规模化生产,以刺龙牙的幼嫩茎段、茎尖为外植体进行组织培养快繁技术研究。结果表明:刺龙牙幼嫩茎段、茎尖先用75%酒精浸泡15s,再用0.1%升汞消毒10min,灭菌效果最好。最佳诱导培养基为MS+6-BA1.0mg·L^-1+GA30.5mg·L^-1+3%蔗糖+0.6%琼脂;最佳继代培养基为MS+6-BA1.5mg·L^-1+IAA0.6mg·L^-1,增殖系数≥4;最佳壮苗培养基为MS+6-BA1.5mg·L^-1+IAA0.6mg·L^-1+GA30.5mg·L^-1;最佳生根培养基为1/4MS+IAA0.7mg·L^-1,生根率90%以上;最佳移栽基质为纯沙,成活率达93%以上。

Abstract:To accelerate propagation speed of Aralia elata, and then promote large-scale production, young stem and shoot tip of A. elata were used as explants for tissue culture technique study. The results indicated that the best protocol for young stem and shoot tip sterilization was 75% alcohol for 15 s, then 0.1% mercury chloride for 10 min. The best induction medium was MS + 6-BA 1.0 mg·L^-1+GA3 0.5 mg·L^-1+3% sucrose + 0.6% agar;the optimum subculture medium was MS + 6-BA 1.5 mg·L^-1 + IAA 0.6 mg·L^-1, with increment coefficient ≥4;the optimal seedling medium was MS+6- BA 1.5 mg·L^-1 +IAA 0.6 mg·L^-1 +GA3 0.5 mg·L^-1;the best rooting medium was 1/4MS+IAA 0.7 mg·L^-1, rooting rate was more than 90%;the suitable transplanting substrate was pure sand, with survival rate more than 93%.
