六堡茶茶褐素的提取工艺及其抗氧化活性研究

目的]为六堡茶茶褐素的提取及生物活性的测定与开发利用提供理论参考。方法]采用正交试验设计方法考察了料液比、提取温度、提取时间、提取次数对六堡茶茶褐素产率的影响,优化了从六堡茶中提取茶褐素的最优工艺,并对所得茶褐素进行了抗氧化活性分析。结果]试验得出,提取次数和提取温度对六堡茶茶褐素产率的影响达到显著水平,料液比和提取时间影响不显著。六堡茶茶褐素提取的最优工艺条件为:料液比1∶30 g/ml、提取温度100℃、提取时间80 min、提取次数4次,在此条件下茶褐素提取率为22.92%。抗氧化活性试验结果表明,茶褐素对羟自由基清除能力高达74%,并且在低浓度时消除效果明显高于VC;对亚硝基的清除率达73%,与VC差别不大;对超氧自由基的清除率虽较VC差些,但仍可达60%以上。结论]六堡茶茶褐素含量较高,且茶褐素具有非常好的抗氧化活性,在抗氧化应用方面前景良好,也为六堡茶的深度开发奠定了坚实的基础。

Extraction Technology of Theabrownine from Liubao Tea and Its Antioxidant Activity Research

Objective] To provide theoretical reference for theabrownine extraction from Liubao tea,biological activity determination,development and utilization.Method] Orthogonal test was adopted to investigate effects of solid-liquid ratio,extracting temperature,time,times on yield of Liubao tea theabrownine,the technique was optimized,and the antioxidant activity of theabrownine was analyzed.Result] The results showed that effects of extraction times and temperature on theabrownine yield reached significant level,while other two factors were not significant.The optimum extraction conditions were determined as follows: solid-liquid ratio 1∶30,extraction temperature 100 ℃,extraction time 80 min,extraction 4 times.Under this condition,the yield could be up to 22.92%.Results of antioxidant activity research showed that the most effectively scavenging activity of hydroxyl radical could be approached to 74%,stronger than Vitamin C's;And the scavenging activity of Nitroso could be approached to 73%,and there was little difference compared with Vitamin C's;Although the scavenging activity of superoxide radical was no better than Vitamin C's,it also could be approached to 60%.Conclusion] Liubao tea has abundant theabrownine,and theabrownine has strong antioxidant activity,which will lay a solid foundation for deeply development of Liubao tea.
