摘    要:通过单因素试验考察了碳源、氮源、血清及辅酶对副猪嗜血杆菌培养的影响,并在单因素试验的基础上,选取蔗糖、酵母粉、硫酸铵及马血清进行4因素3水平试验,最终确定蔗糖2 g/L、酵母粉2 g/L、硫酸铵2 g/L、马血清5%为最优培养基配方。在10 L发酵罐上进行副猪嗜血杆菌分批补料发酵,对比不同补料方式对副猪嗜血杆菌发酵的影响,结果表明利用恒葡萄糖浓度补料将残糖浓度维持在1 g/L时可取得较好的发酵效果,菌体密度及活菌数目分别为16.8、3.6×109 CFU/mL。

Abstract:The effects of carbon, nitrogen, serum and coenzyme on culture of Haemophilus parasuis were investigated by single factor test, and on the basis of single factor test, the orthogonal design of four factors and three levels was employed to test the concentrations of sugar, yeast powder, ammonium sulfate and horse serum.The results showed that the optimized concentrations were that sucrose was 2 g/L, yeast powder was 2 g/L, ammonium sulfate was 2 g/L, and horse serum was 5%. Comparing the impact of different supplement methods on Haemophilus parasuis fermentation in 10 L fermentor, a good fermentation result was obtained when the residual sugar concentration was maintained at 1 g/L by supplementing constant sugar.The result showed that bacterial density and the number of viable bacteria were 16.8 and 3.6×109 CFU/mL,respectively.
