新疆家蚕抗菌肽可有效抑制掌面革兰氏阴性菌的研究

摘    要:抗菌肽具有抑制细菌的作用,是天然防腐剂和潜在的抗生素替代品。为了解新疆家蚕抗菌肽(CecropinXJ)能否有效抑制手掌表面细菌,本研究采用擦拭法获取手掌表面细菌擦拭液,并用不同浓度CecropinXJ处理手掌擦拭液,通过平板菌落计数法评价CecropinXJ的抑菌效果,结果表明随着CecropinXJ浓度增加,其抑菌能力越好。当CecropinXJ添加到0.4%时,抑菌能力达到商业洗手液水平(P>0.05);进一步对16S rDNA进行测序分析,结果发现手掌表面细菌多为革兰氏阴性菌,添加CecropinXJ后,手掌表面的革兰氏阴性菌生长能有效被抑制。本试验结果表明CecropinXJ可以用作消毒清洁产品进行开发。

Abstract:Antimicrobial peptide (AMP) could kill bacteria, which had been used as a natural preservative and potential alternative to antibiotics. To understand whether the CecropinXJ could effectively inhibit bacteria from palm skin, we used sterile water to swab hands and treated with swabbed liquor by different concentrations of CecropinXJ in this study. The antimicrobial activity was evaluated by plate count method. The results showed that the growth of bacteria derived from palm skin could be effectively inhibited via adding different concentrations of CecropinXJ, and the antimicrobial activity became stronger with increasing the concentrations of CecropinXJ. When the concentration of CecropinXJ was up to 0.4%, its inhibitory capacity could be comparable to commercial hand sanitizer (P>0.05). Further study elucidated that swabbed liquor included most of gram-negative bacteria through 16S rDNA sequence analysis. These findings suggested that CecropinXJ could be used as a novel disinfector or cleanser for application potentially.
