摘    要:研究了影响农杆菌GV1301菌株介导的菊苣遗传转化的几种因素,建立了菊苣高效遗传转化体系,获得了转基因抗性苗.结果表明,采用菊苣叶片,经预培养和共培养2 d,农杆菌浸泡时间为5~7 min,卡那霉素20 mg/L,头孢霉素600 mg/L,最高转化频率达7.1%,经PCR检测,目的基因已整合到菊苣基因组中.

Abstract:Factors affecting genetic transformation of Cichorium intybus by Agrobacterium tumefaciens were studied and a highly efficient system for genetic transformation of Cichorium intybus was developed. The result indicated that the optimized conditions for transforming Cichorium intybus leaf fragments were pre-culturing explants for 2 d, infecting them by Agrobacterium tumofaciens for 5-8 min, co-culturing for 2 d, and then regenerating on the medium with kanamycin 20 mg/L and cefotaxime 600 mg/L. The transformation frequency was 7.1%. PCR amplification verified that the target gene had been integrated into genome of Cichorium intybus.
