摘    要:掌叶半夏成熟胚在含2,4-D2.0mg/L,BA0.5mg/L的MS培养基上可诱导形成颗粒状胚性愈伤组织。用附加2,4-D 2.0mg/L,BA0.1 mg/L,CH300mg/L的MS液体培养基对胚性愈伤组织进行悬浮振荡培养,经3 ̄4次继代培养即可得到悬浮的单细胞,其细胞多为圆形或椭圆形,具有较强的分裂能力。经测定,悬浮培养过程中细胞生长曲线呈“S”型,培养20天时细胞数量和鲜重达到最大值;随着

Abstract:Granular embryogenic calli were produced from the mature embryos of Pinellia pedatisecta Schott on MS medium with, 2, 4-D 2. 0, BA 0. 5 mg/L. The embryogenic calli were suspended in MS medium with 2, 4-D 2. 0, BA 0. 1, CH 300 mg/L. After 3~4 times of subculture, single cell suspension were obtained, in which most cells were round or ellipsoidal and fissionable. Cell growth process in suspension culture presented a S-shaped curve. With the growth of cells, the content of solable protein in cultured cells increased, pH value of culture medium decreased. First cell groups and then calli formed when the suspended single cells were cultured in solid or liquid medium. When the regenerated calli were transferred to differentiated medlium with NAA and BA, most of them grew as normal plantlets.
