东北荷包猪MC4 R基因的多态性分析

[目的]分析荷包猪MC4 R基因TaqI酶切位点多态性,为了解其遗传特点奠定基础。[方法]利用PCR-RFLP技术研究荷包猪保种群体MC4 R基因型分布情况,并与国内外猪种的MC4 R基因频率进行比较。[结果]不同品系猪的MC4 R基因的Asp298Asn突变分布差别显著。荷包猪MC4 R基因优势基因型为AA,基因型频率为66.67%,而BB基因型未能检出。荷包猪MC4 R基因型与国内梅山猪、民猪和莱芜猪等稍有不同,与国外汉普夏、大白、皮特兰等差别较大,总体显示国内地方猪种298Asp基因为优势基因,而外种猪基因Asp298Asn突变率显著高于我国地方猪种。[结论]该研究为荷包猪资源保存、品种选育和开发利用提供了科学依据。

Polymorphism Analysis on Melanocortin-4 Receptor (MC4R) Gene in Northeast Hebao Pig

[Objective]The aim was to analyze the polymorphism of TaqI restriction site of melanocortin-4-receptor(MC4 R) gene in northeast Hebao pig and lay the foundation for understanding its genetic characteristics.[Method]The distribution condition of the genotype of the conservation population of Hebao pig was studied by PCR-RFLP technique and the gene frequency of MC4 R of the pig species between home and abroad were compared.[Result]The distribution of Asp298Asn mutation of MC4 R gene of pigs in different species had significant difference.The preponderant genotype of MC4 R gene of Hebao pigs was AA and the genotype frequency was 66.67%,but the genotype of BB was not detected.MC4 R genotype of Hebao pig had slight difference with that of some Chinese species such as Meishan pig,Min pig and Laiwu pig,and had large difference with some foreign species such as Hanpshire pig,Large White pig and Pietrain pig.On the whole,it showed that 298Asp gene of the Chinese pig species was preponderant,but Asp298Asn mutation rate of the gene of the foreign pig species was significant higher than that of the Chinese pig species.[Conclusion]The research provided the scientific basis for the resource preservation,variety breeding and the development and utilization of Hebao pig.
