摘    要:Φ60~150μm牛腔前卵泡在有、无促性腺激素存在下,分别添加0、10%、15%、20%牛卵泡液(BFF)的McCoy′s 5 a无血清系统体外培养15 d,测定其雌二醇(E2)分泌量。结果显示:各添加卵泡液组在培养3、6、9、12 d的E2分泌量均显著高于对照组(0组)(P<0.01)。当培养液中有促性腺激素(FSH、LH)存在下,各添加卵泡液组的E2分泌量更高,其中添加10%牛卵泡液组中Φ>100μm腔前卵泡在培养6 d时E2分泌量达最高,为48.96±11.54 pg/mL。表明培养液中添加一定比例牛卵泡液(BFF)可显著刺激体外培养腔前卵泡E2的分泌,促性腺激素存在下更加强了卵泡液对E2分泌的刺激作用。

Abstract:Bovine preantral follicles(about 60~150 min diameter) were cultured in vitro in the McCoys 5a serum-free system supplementing with different doses of bovine follicular fluid(BFF)in presence or absence of gonadotrophins for 15 days.Estradiol(E2) production quantity were determined every 3rd day.Results demonstrated that E2 secretion quantity in each different doses of BFF group was remarkably higher than the comparison group(0 group) in the 3rd,6th,9th and 12th day of culture separately,and it became more high in the presence of gonadot rophins(FSH and LH).The level of oestradiol(E2)arrived the hig hest value(48.24 pg/mL) in the sixth day of culture for follicles with a diameter of 100 m above in the 10% BFF group.These results revealed that the addition of BFF can significiantly promote E2 production of preantral follicles in vitroculture,which was augmented in the presence of gonadotrophins.
