白羽番鸭脂联素基因内含子多态与肉质的关联分析

摘    要:采用PCR-SSCP法对白羽番鸭脂联素基因内含子进行SNP检测,并分析SNP对肉质的遗传效应。结果表明,检测到3种基因型(CC、CT和TT)和2个等位基因(C和T),CT和C分别为优势基因型和优势等位基因。序列比对发现,在内含子内存在1个SNP位点T290C,该位点在白羽番鸭群体中表现为中度多态,处于Hardy-Weinberg平衡状态(P>0.05);最小二乘法分析表明,CC基因型个体的IMF显著高于TT基因型个体(P<0.05),失水率极显著低于TT基因型个体(P<0.01);CT基因型个体的失水率显著低于TT基因型个体(P<0.05)。结果揭示,T290C位点可能对脂肪沉积有显著影响,CC基因型是白羽番鸭肉质较为优良的基因型。

Abstract:The full-length of adiponectin gene intron of White Muscovy was amplified using speci-fic primers designed from Cairina moschata DNA sequence,and the polymorphism were detected by PCR-SSCP method and its association with meat quality were further analyzed.Two alleles(C and T) and 3 genotypes(CC,CT and TT) were observed,among which C and CT were predominant.One SNP(T290C) was found in the intron of adiponectin gene.The T290C loci belongs to moderate polymorphism and is in accordance with Hardy-Weinberg equilibrium(P>0.05).IMF of the birds of genotype CC are significant higher than that of genotype TT(P<0.05).Percentage of chest muscle loss water of the birds with genotype CC and genotype CT are extremely significant or significant lower than that of genotype TT(P<0.01 or P<0.05).These results suggest that the T290C loci has genetic effect for fatty deposition and genotype CC is the helpful genotype of meat quality in White Muscovy.
