天然和加工产生的两种反油酸对人脐动脉内皮细胞损伤的比较研究

摘    要:为了探讨天然反式脂肪酸11t18∶1和工业反式脂肪酸9t18∶1对人脐动脉内皮细胞损伤及脂肪酸组成的影响。本研究在内皮细胞中加入不同浓度的11t18∶1和9t18∶1,观察其对细胞存活率、细胞凋亡的变化;通过气相色谱测定细胞的脂肪酸组成变化,探讨两种反式脂肪酸对细胞损伤作用的异同。结果显示11t18∶1和9t18∶1均降低细胞活性、增加细胞凋亡率,且存在剂量反应关系。浓度为400μmol/L的11t18∶1和9t18∶1处理细胞后,平均存活率分别下降了9%和11%,平均早期凋亡率为64%和70%。两种反式脂肪酸均可降低细胞内单不饱和脂肪酸和多不饱和脂肪酸的含量,总反式脂肪酸含量明显增加且和浓度呈正相关。在细胞内有少量11t18∶1可以转化为9c11t-CLA,可能与11t18∶1对细胞损伤稍弱有关。高浓度(200~400μmol/L)的11t18∶1与9t18∶1对内皮细胞均有损伤作用,9t18∶1损伤程度较高,但差异不明显。

Abstract:The natural trans fatty acids 11t18∶1 and industrial trans fatty acids 9t18∶1 impact on human umbilical artery endothelial cell injury and fatty acid composition.Our study observed different concentrations of 11t18∶1 and 9t18∶1 on changes of cell viability and apoptosis,and detected the fatty acid composition of the different treatment cells to investigate reasons by gas chromatographic.The results showed that 11t18∶1 and 9t18∶1 decreased cell activity,increased apoptosis rate and existed dose-response relationship.The average survival rate of 400 μmol/L group of 11t18∶1 and 9t18∶1 decreased by 9% and 11%.The average early apoptosis rates were 64% and 70%.Two trans-fatty acids reduced the content of MUFA and PUFA,and increased the total TFA content significantly.The reason that 11t18∶1 group wasweaker on cell injury may due to a small amount of 11t18∶1transformed into 9c11t-CLA in cell.There is no significant difference of high concentrations group(200~400 μ mol / L) of 11t18∶1and 9t18∶1on cell injury.
