苦马豆素对小鼠肝脏抗氧化功能的影响

摘    要:目的]探讨苦马豆素对小鼠肝脏抗氧化功能的影响,进一步揭示苦马豆素的毒性作用机理.方法]将64只昆明种小鼠随机分为对照组、试验Ⅰ组、试验Ⅱ组和试验Ⅲ组,将小花棘豆全草粉碎后,按Ⅰ组添加15;(含苦马豆素30 mg/kg)、Ⅱ组添加30;(含苦马豆素60 mg/kg)、Ⅲ组添加45;(含苦马豆素90 mg/kg)的比例制作颗粒饲料,饲喂至典型中毒症状出现.攻毒第14、28、42、63 d每次每组随机采集4只小鼠的肝脏,检测SOD、GSH-Px、CAT、NOS活性及MDA、NEFA、·OH、NO和Glu含量的变化.结果]与正常对照相比,试验组小鼠肝脏SOD、GSH-Px、CAT等抗氧化物酶的活性极显著下降(P<0.01),MDA、NEFA、·OH、NO含量极显著上升(P<0.01),Glu含量极显著下降(P<0.01).结论]苦马豆素对小鼠肝脏抗氧化功能有显著影响,而且具有一定的时间和剂量效应关系,长期低剂量摄入苦马豆素可引起小鼠不同程度的肝损伤.

Abstract:Objective]To investigate the effects of swainsonine(SW) on the antioxydic function of liver in mice,and reveal the toxicity mechanism of SW in future.Method]Sixty - four Kunming mice were divided into 4 groups(control group and experimental group I,II,III),the dried plant of Oxytropis glabra DC was comminuted,and different amounts of the grass powder(15%,30%and 45%,the corresponding SW content was 30,60,90 mg/kg) were mixed with the feeds in the three experimental groups.The mice consumed the forages freely until typical symptoms were observed.Four mice were selected randomly from each group on the 14 th-,28 th,42 nd and 63rd day respectively,and their livers were collected and the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH -Px),catalase(CAT) and nitric oxide synthase (NOS),the contents of maleic dialdehyde(MDA),renal free fatty acids(NEFA),hydroxyl radical(? OH),nitric oxide(NO) and glutamic acid(Glu),etc.are tested.Result]The results demonstrated that the activity of SOD,GSH - Px and CAT in experimental groups were significandy lower than those of control groups(P < 0.01),and the contents of MDA,NEFA,? OH,NO in experimental groups were significandy higher than those of the control groups(P < 0.01),the content of Glu was significandy lower than that of the control groups(P < 0.01).Conclusion]The results show that SW has remarkable effects on the antioxydic function of livers in mice,and furthermore a time - effect as well as a dose - effect relationship was observed. It is concluded that receiving a low - dose of SW for a long time causes damages in livers of the mice.
