秋水仙素处理对当归愈伤组织生长的影响

为诱导多倍体当归植株，对其愈伤性组织进行了秋水仙素处理，浓度分别为２０、７０、１００和１５０ｍｇ／Ｌ时间为１０、２０、２６和３５ｄ，试验符合二因素交叉分组设计，处理组合１６个，共计接种愈伤组织８００块。结果表明：（１）秋水仙素处理时间对当归治愈组织生长的影响明显大于处理浓度的效应，但处理浓度对当归愈伤组织存在阈值，（２）秋水仙素处理浓度及时间对当归愈伤组织的生长存互作效应，随处理浓度及时间同步递增

THE EFFECT OF COLCHICINE TREATMENT ON THE CALLUS GROWTH OF ANGELICA SINENSIS

Totally 800 callus,being grouped in 16 combinations, of A. sinensis were treated withcolchicine in concentrations of 20,70,100 and 150mg/L for 10,20,26 and 35 days to induce chro-mosomal doubling. The results showed that:(1)the effects of treat duration on the callus growthwere more remarkable than those of the treat concentration, but there would be a high concentra-tion limitation during the colcHnlne treatment to the callus of A. sinensis.(2)there were interac-tions of effect of the treat concentrations and duration on the callus growth, and the death ratesgrew up significantly as the simuItaneous increases of both the concentratiOn and duration. Ofwhich, the combinations of high concentrations together with the shorter duration could not re-suIt in the negative effects on the callus growth. It was suggested that the optimum conditions ofcolchicine treatment for inducing the tetraploidy plants from A. sinensis should be the combina-tions of high concentration together with shorter duration.
