摘    要:采用KromasilC18柱(4.6mm×250mm,5μm),流动相为乙腈-0.1%磷酸(12:88,v/v),流速为1.1mL/min,紫外检测波为334nm,柱温25℃,建立了加味白头翁颗粒剂中秦皮乙素的反相高效液相层析含量测定方法。在所选定的色谱条件下,秦皮乙素各组与辅料、溶剂及复方提取物中其他成分的峰分离良好,秦皮乙素在0.2160-1.2960μg范围内呈良好的线性关系(r=0.9997,n=6),平均回收率为98.27%,RSD为1.16%(n=6)。该方法简便、灵敏、准确,可用于加味白头翁颗粒剂的质量控制。

Content determination of esculetin in modified Radix Pulsatillae granule by RP-HPLC

Abstract:HPLC method was used to establish a simple RP-HPLC method for the content determination of esculetin in modified Radix Puls(aillae granule. The HPLC system was consisted of Tuve Kromasil C18 column (4.6 mm×250mm, 5 μm )and acetonitrile-0.1% phosphoric acid ( 12:88, v/v )mixture as the mobile phase with a flow rate of 1.1 mL·min-1 .The detection wavelength was 334 nm. The column temperature was maintained at 25℃. The excipients and solvents in the granule could be well separated from the drug under such a designated chromatogram condition and (lid not interfere with the assay.A good linear relationship was found between peak area and the concentration of esculetin in the range of 21.60-129.60μg·mL-1( r=0.999 7, n=6), the average recovery was 98.27% and RSD was 1.16% (n=6). The established method was specific, accurate, sensitive and applicable. It can he used to control the quality of modified Radix Pulsatillae granule.
