摘    要:根据Genbank中鸡传染性支气管炎病毒(IBV)核蛋白(NP)基因序列,设计了1对引物,通过鸡胚繁殖IBV,纯化鸡胚尿囊液中的IBV,提取病毒RNA,建立了检测传染性支气管炎病毒的RT-PCR方法。对IBV各毒株(M41,H52,H120,Aust-T以及野毒株等)检测,结果均为阳性,而对鸡的其他病毒性疾病病毒(IBDV,AIV,NDV)进行检测,结果均为阴性。初步结果表明所建立的RT-PCR技术可用于鸡传染性支气管炎病毒各血清型毒株的检测。

Abstract:One pair of primers to amplify the N gene of infectious bronchitis virus(IBV) was designed and synthesized according to megalign results of published IBV sequences in Genbank, and IBV were cultured chicken embryo, RT- PCR technique to detect IBV was established after selecting the best reaction conditions. Although the positive results were obtained from IBV M41,H52,H120,T strains, the negative results were achieved from avian relativity disease virus, IBDV, AIV, NDV. The results showed that the established IBV RT- PCR technique provided a more sensitive, specific and reliable method for diagnosis and epizootic study of the IBV.
