氮源和碳源对高山红景天愈伤组织诱导的影响

摘    要:以高山红景天种子和叶片为外植体,以MS为基本培养基,研究不同氮源含量比例及不同碳源浓度对高山红景天愈伤组织诱导的影响。结果表明,适当调节不同氮源比例可以提高愈伤组织诱导率,而且与适宜浓度的植物生长调节剂配合使用效果更明显;叶片为外植体时,高山红景天雌株和雄株叶片均在硝酸铵和硝酸钾含量为500/3050(mg/L)的培养基上愈伤组织诱导效果最好。糖的种类和浓度也影响诱导率,蔗糖的效果最好,其浓度因材料不同而有所不同,种子和雌株叶片在浓度为3%时诱导率最好,雄株叶片在3%~7%时都能取得好的效果。

Abstract:Took the Rhodiola sachalinensis seed and left as explants and MS as the basic culture medium, change the content of different nitrogen change ratio and carbon source and find the effect to the concentra- tion on callus induction of Rhodiola sachalinensis. The results showed that the induction rate of callus can improve the appropriate regulation of nitrogen ratio, and hormone concentration suitable with the use of the effect is more obvious,leaf as explants,the female and male plants of Rhodiola sachalinensis blades in ammonium nitrate and potassium nitrate content is 500/3 050 mg/L cultured on callus induction effect the best. The kind and concentration of sugar also affected the induction rate, sucrose, glucose and sugar, su crose has the best effect of the three. It varies when its concentration is different, seeds and left are the best when the concentration is 3% ,male plant left can achieve good results when the concentration is 3%~7%.
