软骨藻酸快速检测方法技术研究

摘    要:为比较研究海产品中软骨藻酸日常分析的快速检测技术,筛选出适合不同检测目的的检测技术。本文通过对直接ELISA法、间接ELISA法、胶体金免疫层析法和高效液相色谱法的检测时间、检出限、保质期等进行研究,并对贝类样品中软骨藻酸含量进行测定。结果表明,间接ELISA法的检出限为18ng/L,检测时间为2.5h,在4℃条件下可保存7d;直接ELISA法的检出限为3.58ng/L,检测时间为1.5h,在4℃条件下可保存7d;免疫胶体金试纸条检出限为20ng/L,检测时间15min,在4℃条件下可保存6个月;高效液相色谱法(HPLC)检出限为0.25ng/L,检测时间为6min。从而可以推断这4种检测技术均能达到各国水产品条例软骨藻酸20μg/g限值的要求,而高效液相色谱法检出限最低,免疫胶体金试纸条具有更短的检测时间和较强的稳定性。

Abstract:In order to compare the difference of fast detection techniques for seafood DA,and screen the detection technique for different testing purposes,the detection time,detection limit,quality guarantee of the methods of direct ELISA,indirect ELISA,colloidal gold immune-chromatography for detecting shellfish samples DA were investigated.The results showed that detection limit,detection time and shelf life of different methods were as follows:indirect ELISA with 18 ng/L,2.5 h,7 days at 4 ℃,direct ELISA with 3.58 ng/L,1.5 h,7 days at 4 ℃,colloidal gold immune-chromatographic test strip with 20 ng/L,15 min,6months at 4 ℃,and HPLC with a 0.25 ng/L and 6 min,respectively.It can be concluded that this four detection technology can reach the 20 μg/g of DA limit requirement of the world aquatic product regulation.The detection limit of HPLC is lowest,and the colloidal gold immune-chromatographic test strip has shorter detection time and stronger stability.
