番茄同源四倍体离体诱导及生理特性分析

摘    要:以番茄品种渝粉109为试验材料,在建立番茄植株高效离体再生体系的基础上,将带绿芽生长点的子叶愈伤组织接种到含有秋水仙素的固体培养基中进行诱导。结果表明:以0.2%秋水仙素处理48 h效果最佳。对番茄四倍体与二倍体植株形态、生理指标比较分析发现:四倍体植株长势强,表现出巨大性|四倍体番茄叶片中叶绿素总量、POD活性、SOD活性和可溶性蛋白含量高于二倍体,MDA含量低于二倍体。

Abstract:In this paper,in viro induction of autotetropolid tomato(Lycopersicon esculentum Mill.)with colchicine was studied on the basis of building regeneration system,the appropriate induction method of tetraploid tomato was culturing cotyledon and hypocotyl callus with buds in medium with 0.2% colchicines for 48 hour.Some indexes of diploid and tetraploid plant were compared and analyzed.The tetraploid plants were found to have stronger growth vigor.Its total chlorophyll contents in tomato leaves and POD activity and SOD activity and soluble protein content were higher than those of diploid.But its MDA content was lower than that of diploid.
