摘 要: | [目的]优化大孔吸附树脂法纯化荔枝壳总黄酮的工艺。[方法]比较AB-8、HPD-600和D101 3种大孔吸附树脂对荔枝壳总黄酮的吸附和解吸效果,并对上柱液的pH、黄酮浓度、上柱液体积和洗脱液乙醇体积分数等条件进行优化。[结果]D101大孔吸附树脂适宜荔枝壳总黄酮的提纯,其最佳工艺条件为上柱液pH 5.0,上柱液浓度4 mg/ml,上柱液体积2.5 BV,洗脱液乙醇体积分数80%,洗脱体积2.0 BV。[结论]经D101大孔吸附树脂分离后,荔枝壳总黄酮含量在83%以上。

Separation and Purification Process of Total Flavones from Litchi Pericarp by Macroporous Absorption Resin |

Abstract: | [Objective] This study aimed to optimize the conditions for purification of total flavones from litchi pericarp by macroporous absorption resin.[Method] The flavones adsorption rate and desorption rate of macroporous absorption resins(AB-8,HPD-600,D101) were compared,and the technological parameters of D101 during the purification process were investigated,including on-column liquid pH,flavoneconcentration,on-column liquid volume and volume fraction of elution solution.[Result] D101 macroporous absorption resin is appropriate for the purification of total flavonoids from litchi pericarp.The optimal technological conditions were:on-column liquid pH=5.0;on-column liquid concentration was 4 mg/ml,with a volume of 2.5 BV;elution solution was 80% ethanol,with a volume of 2.0 BV.[Conclusion] The content of total flavones achieve 83% by using the method. |
