摘    要:目的]筛选产纤维素酶的海洋真菌资源。方法]从杭州湾海域采集海水和海泥(沙)等样品,采用海水PDA培养基进行富集培养,然后在羧甲基纤维素钠培养基上进行真菌分离纯化,采用刚果红染色法筛选产酶菌株,并对其中纤维素酶活性较高的菌株进行了26S rDNA ITS扩增及序列测定。结果]共筛选到72株产纤维素酶的海洋真菌,两株高产菌株的同源序列比对结果分别与Penicilliumfuniculosum和Pseudocercosporella fraxini的同源性高达100%和99%。结论]该研究为纤维素酶高产菌株的改造和定向进化奠定了资源基础。

Screening and Identification of Cellulase Producing Marine Fungi from Hangzhou Bay

Abstract:Objective] The aim was to isolate and identify the cellulase producing marine fungi.Method] The seawater and sea mud(sand) were collected from Hangzhou Bay,PDA seawater medium was applied for enriched culture.The medium containing CMC-Na was applied for isolation and purification of marine fungi.And then the cellulase producing strains were screened by the CMC/Congo Red method.The strains with high cellulase activity were amplified by 26S rDNA ITS and then determined by sequencing.Result] A total of 72 strains of marine fungi which could produce cellulase were screened from Hangzhou Bay.The two high yield cellulase producing strains had 100% and 99% homology with Penicillium funiculosum and Pseudocercosporella fraxini,respectively.Conclusion] The research laid a good foundation for the selection and directed evolution of highly cellulase-producing strains.
