摘    要:目的]建立测定虎杖中多种特征成分含量的分析方法,对不同产地的虎杖药材进行品质评价。方法]建立HPLC色谱法,以十八烷基硅烷键合硅胶为填充剂(C18柱250 mm×4.6 mm,5μm);以乙腈-浓度1%磷酸水溶液按以下梯度洗脱,0~30 min,乙腈20%~38%;30~70 min,乙腈38%~100%;流速为1.0 ml/min;检测波长为284 nm。结果]白藜芦醇苷、白藜芦醇、大黄素苷、大黄素、大黄酚及大黄素甲醚对照品浓度在一定的范围内与峰面积呈良好线性关系,平均回收率在95.00%~105.00%(n=5)。结论]该方法操作简便,结果准确可靠,可同时测定中药虎杖类蒽醌及中芪类成分的含量,为虎杖药材的品质评价提供一种简易、可行、特征性强的分析方法。

Simultaneous Determination of Multiple Typical Ingredients in RHIZOMA POLYGONI CUSPIDATI

Abstract:Objective] To set up an analysis method for simultaneously determining the content of multiple typical ingredients in RHIZOMA POLYGONI CUSPIDATI so as to evaluate the quality of related medicinal materials produced in different areas.Method] The HPLC system was established with reverse-phase C18 Hyperrsil column(250 mm×4.6 mm,5 μm),which was packed with the octadecyl silane chemically bonded silica.The mobile phase consisted of acetonitrile and 1%phosphoric acid water solution.Following the injection of test sample,a gradient elution was initiated at a flow rate of 1.0 ml/min: the ratio of acetonitrile in the mobile phase was from 20% to 38% at 0-30 min,and increased to 38% to 100% at 30-70 min.And the UV detective wavelength was set at 284 nm.Result] Through investigation,polydatin,resverotol,emodin-8-O-β-D-glucopyranosid,emodin,chrysophanol and quistin displayed good linear relations within a certain range.The recovery rate ranged from 95.00% to 105.0%(n=5).Conclusion] The method is simple and accurate,and can simultaneously determine the content of the stilbenes and anthraquinones in RHIZOMA POLYGONI CUSPIDATI,as well as providing a simple,feasible and typical analyzing method for the evaluation on the quality of Chinese medicinal materials RHIZOMA POLYGONI CUSPIDATI.
