发酵产金针菇菌丝中超氧化物歧化酶特性研究

摘    要:以金针菇液体发酵产菌丝为研究材料,用聚丙烯酰胺梯度凝胶电泳及抑制剂处理法,研究超氧化物歧化酶(SOD)的产生方式及其同工酶类型.结果表明,在液体培养过程中,SOD属于胞内酶,仅存在于菌丝中,其活性可被氯仿:乙醇和SDS抑制,但对KCN和H_2O_2不敏感,因此认为金针菇菌丝中仅以Mn-SOD为主。

Abstract:Using liquid fermented Flammulina velutipes mycelia as research material, polyacrylamide gradient gel electrophoresis method and chemicel inhibition treatments were used to study SOD producing method and its isoenzyme type in this paper. The results showed that SOD, as intracellular enzyme, only existed in mycelia during liquid culture period, and that its activity might be inhibited by chloroform-ethanol and SDS, but not KCN and H2O2. So it was considered that the main kind of SOD that existed in Flammulina velutipes mycelia was Mn-SOD.
