摘    要:以市售白色金针菇组织分离得到的菌株SH9为试材,通过多孢子杂交获得抗病白色金针菇菌株JK501,以绿色木霉菌株为指示菌进行抗木霉活性测定,对获得菌株的酶活指标、出菇性状和菌株真实性进行考察,以期获得抗病能力强且农艺性状优良金针菇新菌株。结果表明:选育出的金针菇JK501较亲本SH9菌丝浓密、生长速度快,抗木霉能力提高了3.56倍;漆酶、羧甲基纤维素酶、半纤维素酶和淀粉酶等酶活指标和16种氨基酸含量均显著提升;确定JK501的最佳栽培料配方为棉籽皮42%、麸皮18%、核桃木屑33%、豆饼粉5%、石膏2%;最后通过ITS和ISSR多态性证明JK501是不同于亲本SH9的新菌株。

Abstract:The strain SH9 which was isolated from the commercially available white enoki mushroom tissue was used as parents.A new variety of white Flammulina velutipes JK501 was selected by the method of multi-spore hybridization and the ability of resistance to Trichoderma viride was tested,and then the enzyme activity index,mushroom traits and molecular identification were profomed.The results showed that the myceliume of strain JK501 was denser,and grew faster than SH9,and its activity against T.viride was increased by 3.56 times.The activity of laccase,carboxymethyl cellulase,hemicellulase and amylase and the content of 16 kinds of amino acids were significantly increased.The best cultivation formula for JK501 was No.6 medium,cotton seed peel 42%,bran 18%,walnut sawdust 33%,soybean meal powder 5%,gypsum 2%.Finally,it was proved that JK501 was a new strain different from parental SH9 by ITS and ISSR polymorphism.
