‘京欣一号’父本西瓜组织培养研究

摘    要:为建立简单、快速的西瓜诱导不定芽分化的试验体系,以西瓜‘京欣一号’父本为材料诱导无菌苗,接种4~5 d苗龄的子叶和下胚轴进行组织培养,结果表明:采用子叶为外植体诱导的不定芽数量比下胚轴多;以MS培养基为基本培养基,添加BA 1.0 mg/L和IAA 0.5 mg/L时为最佳不定芽诱导培养基,添加0.1 mg/L KT为最适合的不定芽伸长培养基,添加NAA 0.2 mg/L的1/2MS培养基为最佳生根培养基。

Abstract:Cotyledons and hypocotyls at 4-day seedlings induced by seeds of 'Jing Xin No.1' male parent were used to study regeneration system.The results showed that: cotyledons produced much more shoots than hypocotyls;MS supplemented with BA 1.0 mg/L and IAA 0.5 mg/L was the best shoot regeneration medium;To shoot growth,the MS supplemented with 0.1 mg/L KT medium was better than others;1/2 MS supplemented with NAA 0.2 mg/L was the best rooting medium.
