土壤微生物多样性研究的DGGE/TGGE技术

摘    要:分子生物学技术比传统的培养方法可得到土壤微生物种群多样性更全面的信息。变性梯度凝胶电泳(Denaturing gradient gel electrophoresis, DGGE)和温度梯度凝胶电泳(Temperature gradient gel electrophoresis, TGGE)可分离PCR扩增的DNA片段,已成为研究土壤微生物群落多样性的重要手段。本文综述了DGGE/TGGE技术在土壤微生物多样性研究中的应用进展,分析了该方法的主要影响因素及其优点和存在的问题。

Abstract:Molecular biological technology can be used to research and characterize soil microbes which currently can not be cultured by traditional microbial research methods, and to get more and more complete information about soil microbial community, to reveal soil microbial population structure and genetic diversity more accurately. Denaturing gradient gel electrophoresis (DGGE) and temperature gradient gel electrophoresis (TGGE) techniques had been proven to be powerful tools for analyzing microbial diversity, which can directly separating the PCR amplification clips. In this paper, the influence factors and application aspects, as well as the potentials and the limitations of DGGE/TGGE techniques for microbial diversity analysis were reviewed.
