摘    要:【目的】初步构建洋水仙的组培快繁体系,为洋水仙种球工厂化生产奠定基础。【方法】以洋水仙品种Arkle为试验材料,选用其叶片、花茎及鳞茎的不同部位作为外殖体,以MS、1/2MS和1/4MS为基本培养基,分别添加不同质量浓度的a-萘乙酸(NAA)、吲哚丁酸(IBA)、2,4-二氯苯氧乙酸(2,4-D)、6-苄基腺嘌呤(6-BA)和活性炭(AC),研究不同外植体以及不同培养基对小鳞茎诱导、增殖及生根的影响。【结果】以带鳞茎盘的双鳞片作外植体效果较好,鳞片部位以鳞茎外层鳞片的培养效果较好;较适宜的初代培养基为MS+6-BA3.0mg/L+NAA0.5mg/L+IBA0.2mg/L;较适宜的增殖培养基为MS+6-BA1.5mg/L+NAA0.3mg/L,其诱导率可达668.00%;适宜小鳞茎的生根培养基为1/2MS+NAA0.1mg/L+1g/LAC,其生根率可达到80%。【结论】鳞片分化小鳞茎存在着部位效应;6-BA和NAA组合对小鳞茎的诱导和增殖有良好效果;1/2MS与一定浓度的NAA和AC组合有利于生根。
