青海北山林场桦树根围芽胞杆菌分子鉴定及其拮抗活性分析

摘    要:自青海北山林场桦树根围分离筛选到芽胞杆菌菌株166株,根据菌落特征分为6个类群,选择其中31株菌株纯化保存并进行BOX—PCR及ERIC—PCR指纹图谱分析、16SrDNA及gyrB基因序列分析,鉴定出球形赖氨酸芽胞杆菌Lysinibacillussphaericus12株、解淀粉芽胞杆菌Bacillusamyloliq-uefaciens 5株、韦氏芽胞杆菌Bacillusweihenstephanensis7株、芽胞杆菌Bacillussp.5株、苏云金芽胞杆菌Bacillusthuringiensis2株。通过平板对峙试验筛选得到11株对油菜菌核病原真菌Sclerotiniasclerotiorum具有显著拮抗活性的菌株。以MALDI.TOF.MS质谱分析拮抗菌株QBll的活性成分,可产生脂肽化合物表面活性素和泛革素,表明其拮抗活性与脂肽化合物的合成和分泌有关。

Abstract:One hundred and Sixty-six Bacillus strains were isolated from rhizosphere of Betula platyphylla Suk. in Beishan timberland and were classified into six groups after primary morphological identification. Thirty-one Bacillus isolates were chosen from six groups for polyphasic taxonomy identification including BOX-PCR and ERIC-PCR fingerprints analysis, as well as 16S rDNA and gyrB partial sequence analysis. The results showed that Thirty-one isolates were identified as Lysinibacillus sphaericus (Twelve isolates), Bacillus amyloliquefaciens (five isolates), Bacillus weihenstephanensis (seven isolates), Bacillus sp. (five isolates) and Bacillus thuringiensis (two isolates). Eleven Bacillus isolates were screened which present high antagonistic activity to Sclerotinia sclerotiorum by means of the dual culture method screening. The lipopeptide compound analysis of MALDI-TOF-MS showed that strain QB11 (B.amyloliquefaciens) present distinct antagonistic activity to S.sclerotiorum, produced surfactin and fengycin, indicated that the bio-control efficacy of strains were related to the antifungal lipopeptide compounds produced by strains themselves.
