龙麦19 Gli-A1/Glu-A3位点近等基因系低分子麦谷蛋白亚基(LGW-GS)鉴定

摘    要:为了鉴定小麦品种龙麦19(L-19,2*,7+9,2+12)和一对L-19的Gli-A1位点醇溶蛋白近等基因系L-19-626(2*,7+9,5+10)与L-19-613(2*,7+9,5+10)中与Gli-A1位点紧密连锁的Glu-A3位点低分子麦谷蛋白亚基(LMW-GS),利用一套Glu-A3位点特异低分子麦谷蛋白亚基STS标记对L-19、L-19-613、L-19-626及L-19/L-19-626的F2个体进行PCR扩增。结果表明:在具有醇溶蛋白Gli-A1m的个体中(L-19-626类型),Glu-A3e反应均为阳性,在不具有醇溶蛋白Gli-A1m的个体中(L-19和L-19-613类型),Glu-A3c反应均为阳性。说明编码L-19-626醇溶蛋白的Gli-A1 m基因与编码LMW-GS的Glu-A3e基因紧密连锁,编码L-19和L-19-613的Gli-A1位点醇溶蛋白基因(未命名)与编码LMW-GS的Glu-A3c基因紧密连锁。因此,L-19-626与L-19-613近等基因系品质间的差别很可能是由其LMW-GS Glu-A3e与Glu-A3c基因间的差异引起的。

LMW-GS Identification at Gli-A1/GluA3 Loci in NILs of Wheat Variety Longmai 19

Abstract:In order to identify LMW-GS at Glu-A3 locus linked with Gli-A1 locus in common wheat variety Longmai 19(L-19,HMW-GS 2*,7+9,2+12)and a pair of NILs(L-19-613 and L-19-626)at Gli-A1 locus with genetic background of Longmai 19 with HMW-GS 5+10,the materials that L-19,L-19-626 and L-19-613,F2 progeny of L-19/L-19-626 were assayed by a set STS markers of LMW-GS at Glu-A3 locus.The results showed that Glu-A3e amplified specific band in L-19-626 type materials with gliadin Gli-A1m.in L-19 and L-19-613 type materials without gliadin Gli-A1m,Glu-A3c amplified specific band.These results indicated the Gli-A1m at Gli-A1 locus was linked with LGW-GS Glu-A3e at Glu-A3 locus and the undesignated gliadin at Gli-A1 locus in L-19-613 and L-19 were linked with LGW-GS Glu-A3c at Glu-A3 locus.It was possible that quality difference between NILs L-19-626 and L-19-613 was due to LMW-GS gene difference of Glu-A3e and Glu-A3c at Glu-A3 locus.
